With the aid of a dissecting microscope hearts were manually removed from embryos submerged in phosphate buffered saline (PBS). The out-flow tract region was isolated using fine forceps. The region designated out-flow tract is shown in the organogenic image. As much blood was removed as possible before transfer to Trizol. The out-flow tract of embryos of several litters were collected to obtain enough RNA for SAGE library construction.
