With the aid of a dissecting microscope TS17 embryos were removed from uteri under phosphate buffered saline (PBS). The first branchial arch (see organogenic image) was severed from each embryo using mini-Vannas scissors and was then transferred to Trizol. The first branchial arches of several embryos from multiple litters were pooled together to collect enough RNA for SAGE library construction.
