With the aid of a dissecting microscope hearts were manually removed from embryos submerged in phosphate buffered saline (PBS). The ventricles were isolated using mini-Vannas scissors or fine forceps. The ventricle region is shown in the organogenic image. The outflow tract was removed from the ventricular tissue. As much blood was removed as possible from the ventricles before transfer to Trizol. The left and right ventricles from 10 embryos of several liters was collected to obtain enough RNA for SAGE library construction.
